Several lines of experimental evidence are presented suggesting that the L antigens in low potassium (LK) sheep red cells are associated with separate Na(+)K(+) pump flux is distinct from the action of anti-L(l) on K(+) leak flux, implying that K(+) leak transport sites may not be converted into active pumps by the L antiserum. Treatment of LK red cells with trypsin completely abolished both the stimulation of K(+) pump flux and the enhancement of the rate of ouabain binding brought about by anti- L. That this effect is due to a total destruction of the L(p) determinant associated with the LK pump was evident from the complete failure of anti-L(p) to bind to trypsinized LK red cells. The L(p) antigen can be effectively protected against the trypsin attack by prior incubation with anti-L, indicating that the sites for antibody binding and trypsin action may be closely adjacent at the structural level. Trypsin treatment, however, did not interfere with anti-L(l) reducing ouabain insensitive K(+) leak influx, nor did it prevent binding of anti-L(ly), the hemolytically active L antibody which is probably identical with anti-L(l). The functional independence of the L(p) and L(l) sites was documented by the observation that anti-L(l) still reduced K(+) leak influx in LK cells with experimentally induced high potassium concentrations, at which K(+) pump flux is fully suppressed, whether or not anti-L(p) was binding to the L(p) antigen associated with the LK pump. 相似文献

In a continuation of studies on protein intake and aflatoxin B1 (AFB1) metabolism, weanling rats were fed semipurified diets containing either 20% casein or 5% casein for two weeks to determine the effect of dietary protein level on hepatic microsomal epoxide hydrase activity and AFB1 metabolism in an effort to evaluate the role of protein intake on the formation and degradation of the reactive metabolite of AFB1. Styrene oxide was used as substrate for epoxide hydrase since the hypothetical AFB1 2,3-epoxide (AFB-epox) cannot be synthesized because of its lability. Two groups of animals were fed 20% casein diets; one was fed ad libitum and the second was pair fed to the 5% casein group in order to control the effects of total feed intake. The depression of epoxide hydrase activities caused by the 5% casein diets was approximately equivalent to that previously seen with hepatic microsomal mixed function oxidase (MFO) activities with the identical protocol. Similarly, the metabolism of AFB1 to AFQ1 and AFM1 was depressed by the 5% casein diets, with an increase in the production of chromatographically more polar material. The relationship of the MFO and epoxide hydrase activities to AFB1 metabolism and formation of macromolecular adducts is discussed. 相似文献

Twenty isolates of Penicillium oxalicum produced secalonic acid as their major secondary metabolite. Fermentation conditions were determined for toxin production in grain and liquid media. The 50% lethal dose value for mice ranged from 26.5 to 51.7 mg/kg dependent on animal strain and sex, males being more susceptible than females. Secalonic acid was nontoxic and nonteratogenic to the chicken embryo and exhibited poor antibiotic properties. Its potential role in mycotoxicoses is discussed. 相似文献

Flowering in the long day plant Hordeum vulgare L. var. Wintex barley was enhanced by the addition of far red light to the main light portion of the photoperiod. Far red energy was provided to produce quantum flux ratios (660/730 nm) and phytochrome photoequilibria (Pfr/total phytochrome) equivalent to those reported both beneath a leaf canopy and outside a canopy at twilight. The photoperiodic requirement for long days can be completely eliminated by the addition of far red light. However, both the effect of extending the photoperiod without far red and the addition of far red to 12-hour photoperiods were suboptimal. Maximal stimulation was achieved only when far red was added to continuous light. The duration of the period of maximal apex elongation rate, as well as the reduction of the time required for floral initiation, were saturated by three inductive cycles. When far red energy was provided intermittently during 3 days of continuous light, the ability to respond varied in a circadian manner. This enhancement of flowering by far red appears to be mediated by the “high irradiance response” of phytochrome. 相似文献

Summary High resolution electron microscopy of ultrathin sections confirms the presence of a membrane surrounding the tapetal rods in the cat. Cats depleted of taurine exhibit disruption and disorganization of this membrane, probably the first stage of more severe tapetal degeneration. Histochemical localization of zinc shows it to be present on the periphery of the tapetal rods. The amount of zinc present on the periphery of the tapetal rods of taurine depleted cats was greatly reduced. Taurine in feline tapetum, confirmed by autoradiography and direct measurement, was also greatly reduced in taurine-depleted cats. We conclude that both taurine and zinc are localized on the periphery of the tapetal rods and that they contribute to the stability of the membrane. Rubratoxin B, a lactone-containing bisanhydride metabolite of certain toxigenic molds, inhibited (Na+-K+)-stimulated ATPase activity of mouse brain microsomes in a dose-dependent manner with an estimated IC50 of 6 x 10(-6) M. Hydrolysis of ATP was linear with time and enzyme concentration, with or without rubratoxin in reaction mixtures. Altered pH and activity curves for (Na+-K+)-ATPase demonstrated comparable inhibition by rubratoxin in buffered acidic, neutral, and alkaline pH ranges. Kinetic studies of cationic-substrate activation of (Na+-K+)-ATPase indicated classical competitive inhibition for Na+ and K+. Results also showed competitive inhibition for K+ activated p-nitrophenyl phosphatase as demonstrated by altered binding site parameters without change in the catalytic velocity of dephosphorylation of the enzyme . phosphoryl complex. Noncompetitive inhibition with regards to activation by ATP and p-nitrophenyl phosphate was indicated by altered Vmax values with no change in Km values. Inhibition was partially restored by repeated washings. Preincubation with sulfhydryl agents protected the enzyme from inhibition. Cumulative inhibition studies with rubratoxin and ouabain indicated possible interaction between the two inhibitors of (Na+-K+)-ATPase.
